1. Introduction A spectrophotometer is an analytical instrument used to measure how much light a chemical substance absorbs. It is widely used in chemistry, biochemistry, and pharmaceutical analysis. Key concept: The amount of light absorbed is proportional to the concentration of the absorbing species in solution (Beer-Lambert Law). 2. Principle of Operation The spectrophotometer works on the absorption of light by molecules at specific wavelengths. Light Source: Emits light (UV or visible spectrum). UV range: 200–400 nm Visible range: 400–700 nm Monochromator / Filter: Selects a specific wavelength. Sample Holder (Cuvette): Contains the solution being analyzed. Detector: Measures the intensity
